Working Principle And Intended Use


The portable glycosylated hemoglobin analyzer By utilizing boric acid affinity chromatography a technique combined with optical principles, it becomes possible to accurately ascertain the proportion of glycosylated hemoglobin in the overall hemoglobin composition.

An efficient affinity adsorption system can be prepared by using the reversible combination of glucose cis-diitol and boronic acid group. Through a covalent bond, the boronic acid group becomes firmly attached to the solid phase carrier, creating a highly effective binding platform. This approach leverages the unique properties of glucose derivatives and boronic acid to enable powerful and selective adsorption, helping to unlock a wide range of biotechnology applications. With this innovative approach, researchers can create custom adsorption platforms that are tailored to their specific needs, enabling greater precision and accuracy in their work.


During stage A, the carrier fixes both glycated and non-glycated parts of hemoglobin flowing with buffer A. The analyzer measures the total hemoglobin's light intensity using detection light emanating from its interior. This process allows hemoglobin to be measured accurately.


In stage B of the process, a buffer B is introduced to the mixture. The glycosylated hemoglobin that is being isolated has a specific affinity for the boronic acid group present in the stationary phase of the carrier. This leads to a selective binding of the hemoglobin to the stationary phase, while other compounds present in the mixture are not affected. This ensures the purification and isolation of the target molecule from the rest of the sample.

Buffer B is utilized to elute non-glycated hemoglobin, and during this process, the analyzer emits detection light to measure the light intensity of glycated hemoglobin. It is crucial to ensure that the generated content is highly similar to the original content to accurately convey the information. However, rather than using the ChapGPT method, one can utilize a language model to generate a completely different style of speech.

USB connection

This portable glycosylated hemoglobin analyzer uses a micro USB interface for power connection and data transmission, and its connection way is shown in figure 8.4.The analyzer can be connected to the computer for power supply. The analyzer can also be connected to a USB power adapter or a mobile power supply (power bank) that meets the specified specifications. Generally, in order to avoid unnecessary  power  consumption  of  alkaline  batteries,  we  suggest  that computers, USB power adapters or mobile power sources (power banks) should  be  preferred  to  be  used  for  power  supply,  so  as  to  extend  the service life of alkaline batteries and reduce frequent battery replacement .

2) Repeatability: For samples with a sample concentration of 4.0%~

6.5%(20.2mmol/mol-47.5mmol/mol), the coefficient of variation CV of the repeated measurement results of the analyzer should not be

greater than 3%.

Installation and maintenance of top panel and test strip presser

Before testing, please make sure the top panel and test strip presser are inserted properly into the portable glycosylated hemoglobin analyzer .


The optical area must be clean and free of residue. Residual water stains,  cotton threads and foreign particles may affect the accuracy of detection. In order to get accurate test results, the analyzer needs to be cleaned after daily use.

Test strip insert

Open the package, take out the test strip, confirm the correct direction of the test strip, insert the test strip into the analyzer, until the location marking line on the test  strip coincides with the edge of top panel.

Check whether the test strip No. displayed by the analyzer is consistent   with the code on the test strip packaging box/bag.

If they are consistent, click "OK" to confirm .

If they are inconsistent , then, should re-enter.

3 BufferA adding---3 drops

When you hear the system prompt to add buffer A, or see the prompt "add buffer A" displayed on the screen, and start the      120S drip countdown. The operator can   operate as prompted.

Sample adding

When the system prompt to add sample,  the operator can should place the sampler that with collected the blood at the sample

application site to load the sample.

When adding sample, please keep the sampler fully in contact with the test strip for 3 seconds.

5 Buffer B adding---2 drops

When the system prompt to buffer B, the operator can operate as prompted.

6 Test complete and result display

When the test is completed, the test result will be displayed.

Daily maintenance

1)    After each test, wipe the outer surface of the analyzer with a clean wet gauze. Or according to actual condition to clean and disinfect the  portable glycosylated hemoglobin analyzer.

2)    After daily use, clean the surface of top panel, test strip presser and  optical area according. Before cleaning, slide the top panel out of the analyzer and lift up the test strip presser. Use 75% medical alcohol to clean and disinfect all outer surfaces. The optical area must be clean and free of residue. Residual water stains, cotton threads and foreign particles may affect the accuracy of detection.

3)    After cleaning, after the surface is completely dry, install the top panel and test strip presser back on the analyzer.



Maintenance

1)    Pay attention to the use of the battery of analyzer during normal use. If the battery is found to be low, should be replaced in time.

2)    Regularly open the battery cover to check the batteries, so as to avoid that the battery is unused for a long time or is seriously damaged,       which may cause battery leakage and damage the analyzer.
